چکیده

Cells isolated from skin have wide applications in studies of the pathogenesis skin-related diseases and construction 3D equivalents. This study aimed to isolate keratinocytes, fibroblasts, melanocytes human foreskin characterize purity cell types. Keratinocytes, were by differential trypsinization media selection. The types was characterized based on expression gene markers. assessment marker involved RNA extraction, primer design, quantitative polymerase chain reaction (qPCR) immunocytochemical staining. Our results showed that cocultures keratinocytes fibroblasts dermis, could be separated quick culture Dulbecco’s modified Eagle’s medium. remaining are cultured Epilife Melanocytes epidermis selected changing medium M254 Gene suggested cytokeratin 14 (CK14) is a suitable for elastin (ELN) tyrosinase (TYR) tyrosinase-related proteins 1 2 (TYRP1 TYRP2) markers melanocytes. In conclusion, can same sample selection techniques finding will aid isolation pure various regenerative medicine toxicity studies.

In vitro Co-Culture of Human Skin Keratinocytes and Fibroblasts on a Biocompatible and Biodegradable Scaffold

Background: Extensive full-thickness burns require replacement of both epidermis and dermis. In designing skin replacements, the goal has been to re-create this model and make a product which has both essential components. Methods: In the present study, we developed procedures for establishing confluent, stratified layers of cultured human keratinocytes on the surface of modified collagen-chito...

Comparison of Gene Expression Profiles between Keratinocytes, Melanocytes and Fibroblasts

BACKGROUND The skin has many important functions such as protection, preservation, temperature regulation, and vitamin D synthesis. It is composed of a variety of cell types including keratinocytes, melanocytes and fibroblasts. OBJECTIVE We attempted to compare the gene expression profiles between keratinocytes, melanocytes and fibroblast, using cDNA microarray. METHODS Keratinocytes, melan...

Functional characterization of highly adherent CD34+ keratinocytes isolated from human skin.

Compared to murine models, data on cells responsible for the homeostasis of human epidermis are scarce and often contradictory. Given the conflicting results and the availability of clinical grade protocols to purify CD34 cells from a given tissue, we pursued to phenotypically characterize human epidermal CD34+ population. After magnetic separation of whole skin CD34+ and CD34- cell fractions a...
